Cellartis^® DEF-CS™ 시리즈는 Human pluripotent stem cell (hiPSC, ESC) 배양을 위한 배지로써, 줄기세포의 자연분화, 증식률 감소, 핵형 변화, 분화효율 감소 등과 같은 세포기능 저하 없이 장기간 유지 배양할 수 있다. Cellartis^® DEF-CS™로 배양한 줄기세포는 기존의 줄기세포가 Colony로 배양하는 것과 달리, Monolayer 형태로 배양되며, 아래와 같은 장점을 가지므로 보다 효율적인 줄기세포 연구가 가능하다.

[DEF-CS™를 이용한 Stem Cell 배양형태 비교]

Cellartis^® DEF-CS™ Culture System에서 자란 human iPS cell은 기존 culture system에서 콜로니를 형성하면서 자라는 세포의 성장과 다른 monolsyer형태로 배양된다. 낮은 밀도에서의 세포모양 (morphology)은 분화된 것처럼 보이지만 실제 DEF-CS에서 자란 iPS cell은 높은 미분화 상태를 유지하며 배양되는 것을 확인하였다.

DEF-CS™ 500 Culture System를 사용하여 1 well 당 한 개의 GFP-actin hiPSC를 배양하였다. 배양 24시간 후 (그림 A, B)와 배양 16일 후 (그림 C, D, E, F) 세포 형광 사진을 확인해본 결과, 하나의 세포에서 다수의 세포로 균일하게 확대배양 된 것을 확인할 수 있었다.
