Superb Accuracy & Efficiency

극도로 낮은 error rate로 250,000 bp 당 단, 12개의 error만이 확인 되었다. Taq DNA Polymerase보다 10배정도 높은 fidelity 이다.

짧은 반응 시간

Advantage HD DNA polymerase는 매우 높은 priming 효율로 더 짧은 annealing 시간으로도 더 높은 specificity와 sensitity를 보였다.
또한 Anti-taq antibody에 의해 추가적인 7~10분정도의 predanature 시간이 필요 없기 때문에 더욱 효율적이고 신속하게 반응을 이끌 수 있다. 본 제품 이용시엔 기존 Taq보다 annealing time을 짧게 잡으세요(메뉴얼을 필히 숙지하세요).

Robust Amplification

실험 결과 human genomic DNA는 8.5kb 까지, E. coli genomic DNA는 10kb까지, 그리고 λ DNA는 최대 22 kb까지 증폭할 수 있다. GC rich인 Thermus thermophilus HB8 genomic DNA를 주형으로서 임의에 선택한 8 영역(증폭 사이즈는 각각 약 500 bp)을 PCR 증폭 후, 벡터에 클로닝 해, 각 배열에 대해 복수 클론을 선택하여 그 서열을 확인해, mutation frequency를 확인하였다.
그 결과, Advantage HD DNA Polymerase는 Taq DNA Polymerase와 비교해서 10배 fidelity가 높았으며 기존 pfu계열보다 뛰어난 증폭 효율로 조건 설정이 간편하다.

Figure 1. 뛰어난 fidelity 효소- Advantage HD DNA Polymerase. The error rate (mutation frequency) for Advantage HD Polymerase is lower than that of a number of commonly used DNA polymerases. Eight arbitrarily selected GC-rich regions from Thermus thermophilus HB8 genomic DNA were amplified with Advantage HD Polymerase and other polymerases. PCR products (approximately 500 bp representing each region) were cloned into suitable plasmids. Multiple clones were selected and subjected to sequence analysis to determine the error rate percentage (e.g., 1 error/100,000 bp = 0.001%)
