Terra PCR Direct Polymerase를 이용한 간편한 genotyping

Terra PCR Direct Polymerase Mix을 사용하면 DNA 추출과 정제 과정 없이 혈액, 동·식물 조직이나 파라핀 절편 조직 등으로부터 곧바로 PCR 증폭 반응을 진행할 수 있기 때문에 연구자들의 귀중한 시간과 연구비를 절약할 수 있다. 본 효소는 2 kb까지 목적 DNA를 증폭할 수 있으며 70% 이상의 GC 함량이 높은 유전자 증폭에도 매우 유용하다. 또한 매우 높은 민감도를 갖고 있기 때문에 극소량만 획득할 수 있는 귀중한 시료의 증폭에도 적합하다.
Clontech에서는 기본적인 효소 뿐만 아니라 red dye가 포함되거나 FFPE 시료나 FTA card로부터 증폭할 수 있는 다양한 Terra PCR Direct Polymerase Mix 시리즈를 보유하고 있다

Terra PCR Direct는 혈액, 꼬리나 귀와 같은 생검시료나 식물의 잎 등으로부터 direct PCR하도록 최적화되어 있다. 이때 동물 조직으로부터 곧바로 증폭한 PCR product는 agarose gel에 분석되지 않는 경우가 있는데 이때 gel loading buffer에 Proteinase K를 첨가해서 로딩을 하면 DNA가 well에 걸리지 않아 전기영동으로 쉽게 분석할 수 있다. 따라서 만약 동물 조직으로부터 direct PCR을 하고자 할 때 Terra PCR Direct Polymerase Mix와 함께 Proteinase K를 첨가하는 것을 추천한다.

Terra PCR Direct has been optimized for direct PCR amplification from animal and plant tissues, such as blood (Figure 1), tail and ear biopsies (Figure 2), or leaf cuttings (Figure 3). Importantly, PCR products amplified directly from animal tissues do not always resolve well on agarose gels (Figure 2, Lane 1); however, this can be easily prevented by adding proteinase K to your gel loading buffer (Figure 2, Lane 2). Consequently, we recommend ordering Proteinase K along with Terra PCR Direct Polymerase Mix if you are amplifying directly from animal tissues.

Applications

Perform direct PCR amplification from tissue samples, crude extracts, and dirty templates without having to extract and purify the DNA.
